چکیده

Abstract Background NANOG is a core transcription factor (TF) in embryonic stem cells (ESCs) and primordial germ (PGCs). Regulation of the gene by TFs, epigenetic factors, autoregulatory factors well characterized ESCs, transcriptional regulation established these cells. Although plays key role cells, molecular mechanism underlying its PGCs has not been studied. Therefore, we investigated that regulates chicken ( cNANOG) ESCs. Results We first identified start site cNANOG 5′-rapid amplification cDNA ends PCR analysis. Then, measured promoter activity various 5′ flanking regions ESCs using luciferase reporter assay. expression required regulatory elements, which were positively regulated POU5F3 OCT4) SOX2 negatively TP53 PGCs. The proximal region contains positive element (CCAAT/enhancer-binding protein CEBP )-binding site) Furthermore, small interfering RNA-mediated knockdown demonstrated , played cell type-specific . Conclusions show for time different trans -regulatory elements control manner. This finding might help to elucidate

The effect of BMP4 on mouse embryonic stem cell proliferation and differentiation into primordial germ cells

Background and Aim: Artificial gamete production from stem cells is a novel strategy for treatment of infertility. Among various stem cell sources, embryonic stem cells (ESC) can be considered as an appropriate source for in vitro formation of germ cells. In this study we evaluated the effect of BMP4 on proliferation and differentiation of mouse embryonic stem cells into primordial germ cells (...

A vector-based system for the differentiation of mouse embryonic stem cells toward germ-line cells

Objective(s):To culture thein vitro mouse embryonic stem cells (mESCs) and to direct their  differentiation to germ-line cells; in present study we used a vector backbone containing the fusion construct Stra8-EGFP to select differentiated ES cells that entered meiosis.  Retinoic acid was used to differentiate embryonic stem cells to germ cells. Materials and Methods: A fragment of Stra8 gene pr...

isolation, culture and characterization of chicken primordial germ cells

nowadays, production of recombinant proteins in eukaryotes is gaining good deal of attention. transgenic chicken as a eukaryotic system has a high potential for producing recombinant proteins. post-translational changes, especially glycosylation, are characteristic of the eukaryotic proteins. in practice we need to choose a proper expressing host when considering over-expression of a recombinan...
